Method | TIRF microscopy and epifluorescence. Approximately spherical single and large budded cells with unpolarized patches were scored for each strain. Surface area of individual cells was estimated as an average of sphere surface areas calculated from four diameters (at 0°, 45°, 90°, and 135°) measured from maximum intensity projections. |
