Method Abstract: "Study design and methods: Aliquots of autologous RBCs [red blood cells] from eight healthy adult subjects were labeled separately at four discrete biotin densities, mixed, and infused. The proportion of each population of BioRBCs [biotinylated red blood cells] circulating was determined serially by flow cytometry over 20 weeks. For each population, RCS [RBC survival] was assessed by the following: 1) posttransfusion BioRBC recovery at 24 hours (PTR(24)) 2) time to decrease to 50% of the enrichment at 24 hours (T(50)) and 3) mean potential lifespan (MPL)."
Comments P.1052 right column 2nd paragraph: "Tests for hemolysis of transfused autologous BioRBCs Serial blood samples were drawn before transfusion of BioRBCs (range, 6 to 26 hr) and at 60 minutes and 24 hours after transfusion of BioRBCs to detect acute changes in blood Hb concentration, reticulocyte count, plasma-free Hb, and haptoglobin. In addition, blood Hb and reticulocyte count were measured every 1 to 2 weeks. Comparison of pre- and posttransfusion values did not suggest hemolysis (Tables 1 (BNID 112492) and 2) including Subject 4, despite the rapid removal of RBCs biotinylated at 162 µg/mL."
