"Intermediates of RNAP assembly (immature
RNAPs) have to be taken into account (13), because the RNAP content of cells is determined by measuring the fraction of
total protein mass that is in ß and ß' subunits of RNAP (4). Some
of these subunits are not or are only partially assembled into
functional RNAPs in the cell (primary source). These assembly intermediates
are located in the cytoplasm and radioactively labeled ß and ß' subunits appear in the nucleoid after ˜5 and 2.5 min,
respectively (primary source). The larger of these times is likely to correspond
to the time needed to fully assemble an RNAP plus the
transition to the nucleoid, thereby providing an upper bound
for the maturation time." |
