Method |
Isolation of human brain 6PF2K/Fru-2,6-P2ase isoform cDNAs and gene. Subcloning and mutagenesis. Expression and purification of 6PF2K/Fru-2,6-P2ase. Protein kinase assay. For equations and method used to determine 6PF2K kinetic models and statistics see text. |
Comments |
The 6PF2K (6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase) activity was measured at pH 7.5 with 5 mM ATP and 1–5000 µM Fru-6-P. The Fru-2,6-P2ase activity was measured at pH 7.5 with 50 µM Fru-2,6-P2. |
