DNA per mass was calculated from DNA per cell and cells per mass: GM(genome per mass)=GC(genome per cell)XCM(cell per mass) These calculated values closely agree with direct measurements of DNA per mass (Fig 1 in primary source), using the colorimetric diphenylamine reaction with E. coli DNA as a calibration standard and assuming 1 A260 unit of E. coli DNA to correspond to 2.86X10^13 kbp (for a GC content of 0.5) and the E. coli genome to be 4,700 kbp. Calculated from data in table link in ref and according to average molecular mass of DNA base pair of 618 Da: (618[g per mole base pairs]/Avogadro's constant[number of base pairs per mole])X(genome equivalents per cell)X(number of base pairs per genome)=weight [g/cell]:(618/(6X10^23))X(1.56)X(4.7X10^9)=7.57X10^-15 g/cell |
