Method Each experimental run was initiated by adding one or both labeled proteins in gel buffer at the gel-solution interface using a flexible needle to create a step function of concentration. Initial protein concentration was set at 800 nM for IGF-I. All experiments were run at 24°C, and the results corrected to 25°C using the relation D?/T = constant, where ? is the viscosity of water and T is the absolute temperature. Table 1 shows the values of D obtained by fitting Eq. 4 to the data from each experiment for the two labeled proteins, combining data from both image fields at five time points each.
