Method Morphometric Analysis—HeLa and COS-7 cells expressing RFP fused to a nuclear localization signal were trypsinized, suspended in phosphate-buffered saline, and observed under an inverted microscope. Thirty cells for each were imaged for differential interference contrast and fluorescence to measure the diameters of cells and nuclei, respectively. The volumes of cells and nuclei were calculated assuming that they were spheres. The nuclear and cytoplasmic volumes of HeLa cells were 0.22 and 0.94 pl, respectively. Those of COS-7 were 0.14 and 2.4 pl, respectively.
