Method The yeast is first cultivated for 4 h in a complete Rider medium containing 9 mM Pi and 111 mM glucose as a carbon source. Then the cells are placed on the same medium except depleted of Pi under sterile conditions and the culture is grown during 7 h. Further, the cells are again placed on fresh complete medium, where the culture is growing for 2 and 4 h. The degree of polymerization (n) of polyP fractions was estimated from the areas of 31P-NMR-spectral peaks. As seen from Fig. 2, the polyphosphates of the acid-soluble fraction can be separated into two sub-fractions by precipitation with barium salt first at pH 4.5 and then at pH 8.2.
