Method in vitro, single molecule measurements. P.621 right column bottom paragraph: "The 5' end of one strand of a 48.5 kilobase (kb)-long duplex lambda phage DNA molecule is attached to the bottom surface of a glass flow cell using a biotin–streptavidin linker. The opposite 3' end bears a digoxigenin moiety and is linked to a bead that is 2.8 µm in diameter and coated with anti-digoxigenin antibody (Fig. 2a, b)."

These values are in agreement with those observed in a previously published single-molecule experiment [Wuite et al, PMID 10716452, that gives polymerization rates of over 100 bases/sec for T7 DNA] and ensemble studies [PMID 1846298, PMID 3316214]."
