P.72 left column bottom paragraph: "The properties of all test cellulosic substrates are summarized in Table 1 [primary sources]. The enzymatic hydrolysis tests were conducted with 100 mL of total reaction volume in Erlenmeyer flasks. The enzyme solutions of EglS (50 U), Cel (50 FPU), and the mixture of EglS and Cel (50 U and 50 FPU, respectively) were loaded into separately into flask containing 0.5 g of each dried substrate in the 50 mM sodium citrate buffer (pH 4.8). Moreover, 0.01% sodium azide reagent was added to prevent microbial contamination. For controls, all the studied substrates were suspended in the buffer solution without any added enzymes. All flasks were incubated at 50°C in a shaking incubator (180 rpm). The samples were collected every 20 h for 100 h and boiled for 5 min to stop the hydrolytic reaction. Subsequently, the samples were centrifuged and filtered through a 0.22-μm filter for sugar analysis by HPLC [High-performance liquid chromatography]. The initial and overall sugar production rates of each hydrolytic reaction were calculated from the total concentration of glucose, cellobiose, and xylose produced in the first 20 h and the entire range of 100 h, respectively." |
