P.3 right column bottom paragraph: "Ratiometric pHluorin [ref 21] was expressed in E. coli. Ratiometric pHluorin is a GFP-based sensor whose 405ex/480ex ratio changes in response to changes in the pH, as confirmed by collapsing the ΔpH and changing the intracellular pH. Cells expressing ratiometric pHluorin were cultured under continuous culture conditions and observed under a microscope. One major peak was observed around the ratio 1.9 and one minor peak was observed around the ratio 1. The major peak was fitted by the normal distribution." |
Comments |
P.4 left column: "If the distribution observed in this condition is assumed to be the same as the experimental noise in the major peak, the true distribution of the major peak would be estimated to be N (1.90, 0.076), which corresponds to a pH value of 7.98±0.18 (mean±SD) (See Supplementary Methods)." There is another, much smaller peak to the bimodal distribution around the ratio of 1.0, corresponding to a pH ~= 6.5. |
