Pyrophosphatase activity in the cell extract was measured
in a 0.1-ml assay mixture containing 25 mM Tris hydrochloride
(pH 7.8), 20 mM KCl, 2.5 mM MgCl2, 0.1 mM PPi,
100,000 dpm of 32PP, and appropriately diluted crude extract
solution. |

During growth,
cellular PPi remained nearly constant, between 0.9 and 1.3
mM. It is noteworthy that this concentration is identical to
that in the parental strain K37. |
