"Chloroplast DNA in Chlamydomonas, both in vegetative cells and in zygotes, occurs as aggregates (‘nucleoids’) of variable shape and size, sometimes up to 2.5µm in their longest dimension, scattered between the thylakoids of the single large cup-shaped plastid (primary sources). They can be seen easily by fluorescence microscopy after staining with such DNA-binding fluorochromes as 4’,6 diamidino-2-phenolindole (DAPI), mithramycin or ethidium bromide (EB) (after RNase treatment)." |
