Method "Pieces of tissue were macerated on a slide in 0.1 M NaEDTA and viewed with a Nikon (Tokyo, Japan) Optiphot microscope using Nomarski differential interference optics. Individual mesophyll cells were quickly separated on maceration and all of the chloroplasts within each cell could be counted easily."
Comments "Mean values for cell plan areas derived from 90 cells were obtained for each genotype and inserted into the regression equation to obtain mean values for chloroplast number per cell. These values and their standard deviations are given in Table 3 and Fig.1 illustrates the variation in mean chloroplast number within and between ploidy levels." See note above table
