Method |
High-density oligonucleotide arrays from Affymetrix
were used to study the degradation of RNA over essentially
the entire transcriptome of E. coli MG1655. After addition of rifampicin, which prevents initiation of
new transcripts by binding to the ß subunit of RNA polymerase
(Campbell 2001 PMID 11290327), the total intensity for all mRNAs
decreases exponentially with time (R = 0.98) with an estimated
overall chemical half-life of 6.8 min. |

agreement with a reported half-life of 7.5 min for total pulselabeled
RNA in comparable conditions (Mohanty and Kushner
1999 PMID 10594833) |
