Method | "DNA from isolate S0385 was sequenced using pyrophosphate sequencing technology to 16 fold coverage by the 454 group of Roche Applied Sciences. Resulting reads were assembled into contigs and were placed into the correct order and direction using the USA300 FR3757 genome as a scaffold. All gaps between contigs were closed by conventional PCR using primers based on the sequence of flanking contigs followed by sequencing of the products." |
