Method |
Extraction and Assay of Polyphosphatase. The slime mold was harvested in 0.05 35 Tris, pH 7.6, containing 2 m35 MgCl2 and 1 mM beta-mercaptoethanol. The cell suspensions were used immediately or frozen at-70degrees and stored at -20degrees the time of assay. Homogenization was performed in a French pressure cell at 1400 kg/cm2. |
