Reduction in adenylate cyclase activity in crude membrane of CHO due to low CDR levels

Range 30-40 %
Organism Chinese hamster ovary (CHO)
Reference Evain D, Klee C, Anderson WB. Chinese hamster ovary cell population density affects intracellular concentrations of calcium-dependent regulator and ability of regulator to inhibit adenylate cyclase activity. Proc Natl Acad Sci U S A. 1979 Aug76(8):3962-6.PubMed ID226991
Method Cells grown on dishes were washed, harvested, and homogenized, and crude membranes were isolated. Cyclic AMP phosphodiesterase was assayed by measuring the formation of [3H]AMP from cyclic [3H]AMP. CDR was assayed by its ability to activate CDR-dependent cyclic AMP phosphodiesterase purified by affinity chromatography with CDR coupled to Sepharose
Comments This inhibitory effect occurred at low concentrations (6-600 ng/ml) of calcium-dependent regulator (CDR). The effect was lost at concentrations of CDR above 600 ng/ml.
