Method |
"The decay of a given mRNA may be probed in
at least three ways: (1) through correlations
between steady state amounts and synthesis rates
(2) through the enzyme induction kinetics in the
case of inducible enzymes or (3) through the disappearance
of its hybridization signal after a stop
of transcript initiation by rifampicin. Here [researchers] have
employed all three methods. This has allowed [them]
to analyze the influence of rifampicin on the functional
and chemical decay of lacZ and rplN mRNA
sequences from the wild-type lac and spc operons,
respectively, and from an artificial Pspc-rplN- lacZ
fusion operon." |
