Method In order to measure the pH of E. coli, researchers devised a procedure based on fluorimetry of plasmid-expressed GFP mutant 3* (GFPmut3*) and YFP gene fusions.
Comments After HCl addition, the pHs observed both for cytoplasm (Fig. 8A) and periplasm (Fig. 8B) fell to just below pH 6.0. The cultures with periplasmic GFPmut3*, showed minimal recovery of the pH (Fig. 8B).
