Method "Cells of all strains were frozen in 0.6M sorbitol with 10mM CaCl2 and 10mM Hepes, pH 7.5. Cell concentrations during the electric pulse were 1?10^9cells/ml and 2?10^9cells/ml for non-frozen cells and cells after freezing with calcium, respectively. The electric pulse settings (for 25µF, 200O) were 10.0kV/cm for Sch. pombe and 11.0kV/cm for Saccharomyces cerevisiae, for both non-frozen cells and cells after freezing."
