Method To obtain a single molecule map of yeast nucleosomes, researchers isolated mononucleosomes from eight independent biological replicates and fully sequenced ~503,000 of the nucleosome DNAs, using a parallel sequencing technology whose sequence reads are ~200 bp long.
Comments several studies characterized the nucleosomes' intrinsic sequence preferences primarily by ~10 bp periodicities of specific dinucleotides along the nucleosome length, thought to facilitate the sharp bending of DNA around the nucleosome.
