E. coli cells were cut into 65 nm thick serial ultrathin sections after rapid cryofixation and freeze-substitution. In addition to the structome analysis by serial ultrathin sectioning, PCM (phase contrast microscopy) and SEM (scanning electron microscopy) examinations were performed on the same bacterial sample because the fundamental morphological studies were achieved with these observation techniques." |

Data obtained from three analysis methods were compared. Table.1 shows the results of one-dimensional analysis examined by PCM, SEM and serial ultrathin sections with transmission electron microscopy (TEM)." P.287 left column bottom paragraph: "However, in the result obtained from SEM measurement, average cell diameter was 0.49 μm with the smallest SD of 0.03 and was significantly smaller than those of PCM and structome analysis based on examination of serial ultrathin sections (both P < 0.001). Average diameter of cytoplasm at PM was 0.85 ± 0.06 μm measured with only structome analysis based on serial ultrathin sections because PM cannot be visualized with SEM and PCM analysis (Table 1)." |
