Abstract: "To determine their shape and volume, cells were separated from the sediment matrix by multi-layer density centrifugation and visualized via epifluorescence microscopy (FM) and scanning electron microscopy (SEM). Total cell-carbon was calculated from amino acid-carbon, which was analyzed by high-performance liquid chromatography (HPLC) after cells had been purified by fluorescence-activated cell sorting (FACS)." P.4 right column top paragraph: "For example, the mean cell volume of filtered E. coli cells was 0.68 μm^3, while that of non-filtered E. coli cells was 0.76 μm^3. The cell volumes of filtered elongated cells (rods and filaments) from the sediment samples were therefore corrected by the factor 0.76/0.68 = 1.13 (Table 1)." |
