Method |
Abstract:"Here [investigators] report a reflected light-sheet superresolution microscopy method capable of imaging inside the mammalian nucleus with superior signal-to-background ratio as well as molecular counting with single-copy accuracy. Using reflected light-sheet superresolution microscopy, [they] probed the spatial organization of transcription by RNA polymerase II (RNAP II) molecules and quantified their global extent of clustering inside the mammalian nucleus." |
Comments |
P.684 left column 2nd paragraph:"Moreover, the density of RNAP II molecules observed was found to be 37.2 ± 4.1 molecules per μm^3 (n = 12,482 molecules in eight cells), which translates to a total of 80,200 ± 8,800 RNAP II molecules in the entire nucleus (Materials and Methods), in good agreement with values reported in previous studies (refs 13, 15, 37). Therefore, [investigators] conclude that [they] indeed exhaustively imaged most of the labeled RNAP II molecules in the optical section illuminated." |
