Method |
"Cells of E. coli were loaded with the membrane-permeable acetoxymethyl ester of fura-2, which was cleaved intracellularly to give the free pentaacid." |
Comments |
p.143 left column 4th paragraph:"Different channels exhibit a unique conductance that should reflect a range of pore diameters ranging from the size of a hydrated potassium ion (~6 Å E. coli YnaI) to MscL (large-conductance Mechano sensitive channels, ~30 Å diameter). Such pores allow the rapid release of osmotically-active solutes from the cytoplasm in a manner that is limited predominantly by the diameter of the pore [ref 70]. The pores are lined with hydrophobic residues and some of these residues also form the hydrophobic seal that maintains a non-conducting pore when the channel is ‘closed’ [ref 27], [ref 71] and [ref 72]." |
