"In simulations using dissociation of severed oligomers
and an estimate of ~40µM cytoplasmic cofilin measured on
immunoblots (V. Sirotkin, unpublished observation), actin
patches disassembled as quickly as in cells, if the rate constant
for subunit loss by severing kChop [rate constant for severing] was 0.003µM^-1×sec^-1
(Figure 2B). Thus the modeling and simulations show that depolymerization alone cannot account for the turnover of
actin and associated proteins, but severing and fragment
dissociation is a reasonable alternative hypothesis consistent
with the experimental measurements." |
