Method	Total amount of cellular RNAs (pg per cell) was determined for three independent preparations of S288C cells grown in YPD medium [(1%(w/v) yeast extract/2%(w/v) Bacto peptone/ 2%(w/v) glucose)] using a modified hot-phenol method [PMID 8643554] and NaOH/PCA[perchloric acid] method [PMID 1097403].
