"An apparent dissociation constant (KD) of ~100 nM was estimated
from the in-line probing data generated in the presence of
roseoflavin (Fig. 1C and D). Compared to the KD values previously established for the 165 ribD RNA (refs 5,10), the aptamer has a stronger
affinity for FMN (KD ~5 nM) than roseoflavin, whereas riboflavin
(KD ~3 µM) binds less tightly than the antimetabolite (Fig. 1D).
These findings suggest that the dimethylamino group on the flavin
ring of roseoflavin (Fig. 1B) permits more favorable binding to the
aptamer than the methyl group it replaces." |
