Method |
"Most observations and measurements of starch
granules in leaves have been made on a transmission
electron microscope (TEM) or light microscope images
of sections through mesophyll cells." And flow cytometry |
Comments |
"[Researchers] developed two methods for estimating starch
granule numbers per chloroplast. In method 1, good
resolution of granules within chloroplasts is obtained by
imaging the surface of a resin block containing leaf
material in a SEM...Method 2 involves preparation of intact chloroplasts
from a leaf sample, measurement of the starch content
of a known number of chloroplasts, then estimation of
the average volume of starch granules in preparations
of starch from replicate leaf samples." |
