"Pieces of tissue were macerated on a slide in 0.1 M NaEDTA and viewed with a Nikon (Tokyo, Japan) Optiphot microscope using Nomarski differential interference optics. Individual mesophyll cells were quickly separated on maceration and all of the chloroplasts within each cell could be counted easily." |
