“Sucrose gradients can be used for the rate-zonal fractionation of macromolecules and macromolecular complexes (e.g. nucleic acids, ribosomes and polysomes). Sucrose gradients are used not only for the rate-zonal fractionation of many types of particles, but also for the isopycnic separation of viruses, cell organelles and , where viability is not essential, of cells also. Table 3 indicates the buoyant densities observed for some such samples. The gradients can be unloaded by any of the standard methods and the shape of the gradient can be readily determined from the refractive indices of the fractions.” |
