In order to measure the pH of E. coli, researchers devised a procedure
based on fluorimetry of plasmid-expressed GFP mutant
3* (GFPmut3*) and YFP gene fusions. |

After HCl addition,
the pHs observed both for cytoplasm (Fig. 8A) and periplasm
(Fig. 8B) fell to just below pH 6.0. The cultures with periplasmic
GFPmut3*, showed minimal recovery of the pH (Fig.
8B). |
