Free intracellular calcium was determined from fura-2 fluorescence and using the following equation: [Ca2+]i= Kd(F - Fmin)/(Fmax - F) where F is the fluorescence intensity of dye in the cells, Fmax and Fmin
are the intensities at saturating and zero calcium concentrations,
respectively, and Kd is the dissociation constant for fura-2 determined
under the appropriate conditions. |
