Method |
Abortive Initiation Assays-In the absence of a complete
complement of nucleoside triphosphates, RNA polymerase
synthesizes the tetranucleotide ApApUpU at the UV5 promoter. For
the activity assays, the number of moles to ApApUpU produced
per mole of promoter was determined as a function of
the reaction time t under three different conditions. KB and kf were obtained from Tau (average time necessary for open complex formation) plots |
Comments |
The forward and reverse kinetics of open complex formation between Escherichia coli RNA polymerase and the lac UV5 promoter have been studied in the temperature range of 15-42°C. 0.012sec^-1 for 19°C. 0.11sec^-1 for 45°C |
