Method |
Two clones of recombinant Escherichia coli BL21
strain from a LB agar plate were inoculated into
two 300 ml flasks containing 50 ml LB media
and incubated at 32 °C with shaking at 200 rpm
for 10-12 h. Batch and fed-batch cultivations were carried out
in an in situ autoclaved fermenter (volume of
12.8 l, Bioengineering Co. Switzerland) with 6 l
initial media and 500 ml inoculum at 32 Batch and fed-batch cultivations were carried out

initial media and 500 ml inoculum at 32 °C with
stirring at 300 rpm and 18 l/min aeration rate. Cell growth was measured turbidimetrically at
600 nm and converted to dry cell weight (DCW)
by an appropriate calibration. The broth sample
was centrifuged, washed with distilled water for
three times, and dried in an oven at 105 °C until
it became constant. |
Comments |
Although the final biomass (around 69 g/L)
was almost the same in all fed-batch cultures, the highest product concentration (13.6 g/l) was achieved at
the specific growth rate of 0.15 1/h and the lowest (9.6 g/l) at 0.25 1/h. |
