Method |
When 100 micrograms/ml erythromycin is added to nutrient agar plates, it is possible to obtain a 30-fold enrichment for cds mutants from a mutagen-treated stock, as judged by colony autoradiography. Using this approach researchers isolated 38 new cds mutants, nine of which are unable to grow at a culture pH greater than 8 |
Comments |
The quantitative phospholipid compositions of GN85
(cds+), GN80 (cds-8), and GN80-8 (a spontaneous pH-resistant
revertant of GN80) were determined in samples taken
immediately before and about 2 h after the pH shift (Table
link). The compositions of GN85 and GN80-8 were similar-
neither accumulated significant amounts of phosphatidic acid,
and both contained slightly more phosphatidylglycerol at pH
8.5. |
