Utility of the housekeeping genes 18S rRNA, beta-actin and glyceraldehyde-3-phosphate-dehydrogenase for normalization in real-time quantitative reverse transcriptase-polymerase chain reaction analysis of gene expression in human T lymphocytes. Method |
Levels of 18S rRNA, GAPDH mRNA and b-actin mRNA are expressed as units/ml as defined from triplicate serial dilutions of a pool of total RNA extracted from anti- CD3 MoAb OKT3-stimulated PBMCs. One unit was defined as the signal obtained by 10 pg of this RNA pool. All qRT-PCR assays for different RNA species were run separately. |
Comments |
n=6. Expression levels of 18S rRNA were determined in freshly isolated jejunal intraepithelial and lamina propria lymphocytes. The expression levels were almost identical to that of freshly isolated PBMCs. Note that copy number is per 100 cells |
