Method |
ATP was extracted from frozen tissue powder with ice-cold 10 % perchloric acid (v/v) with 5 mM EDTA. The extract was centrifuged at 18 500 g for 10 min at 4 °C and the supernatant was collected. KOH at 3 M was then added to the supernatant to adjust the pH to 7–8. After the pH adjustment, the solution was centrifuged at 18 500 g for 10 min at 4 °C, and the supernatant was stored at –80 °C until assay. |
