Method P.509 left column 3rd paragraph: "Mitochondria were isolated from cell suspensions according to the "no gradient" method of Bogenhagen and Clayton (Bogenhagen and Clayton, 1974)." P.509 left column bottom paragraph: "Quantitation of mtDNA: Mitochondrial DNA (mt DNA) was measured using the method of Tapper and Clayton (Murphy et al., 1984)." P.509 right column 2nd paragraph: "Mitochondrial counting: Isolated mitochondria from all cell types were stained with Rhodamine 123 according to the method of Johnson, Walsh, and Chen (Johnson et al., 1980)." P.509 right column 3rd paragraph: "Enzyme assays: Cytochrome aa3 was measured spectrophotometrically by the method of Simon et al. (1973)." P.509 right column 4th paragraph: "Malate dehydrogenase: Malate dehydrogenase was measured spectrophotometrically by the method of Rudolph, Baugher, and Beissner (Rudolph et al., 1977)."
Comments P.511 left column 2nd paragraph: "Table 5 summarizes the data concerning various parameters of mitochondrial composition and number. Mt DNA/mt (or per mitochondrial fragment) shows almost no variation between values in any single cell type, between different cell types, and between cells from various mammalian species. The low standard error in a given cell type could be accounted for by the existence of numerous individual mitochondria or by fragments derived from a single giant mitochondrion. The essentially identical values in the various cell types from the four species would require that either individual mitochondria from mammalian species have identical levels of mt DNA or that giant single mitochondria from various mammalian species have identical levels of mt DNA. This finding would be difficult to reconcile with the finding that total cell mt DNA varies in various cell types and species. It, therefore, appears likely that based on these five cell types, mammalian mitochondria contain 2.6 mt DNA molecules/mitochondrion." P.511 left column bottom paragraph: "The number of mt DNA molecules per cell shows little variation in any cell type but varies widely from one cell type to another."
