Method |
P.2808 right column 2nd paragraph: "To Measure the transcription elongation rates, the cultures were induced by addition of IPTG [Isopropyl ß-D-1-thiogalactopyranoside] (1mM) at an optical density at 436 nm of 0.5, and the time elapsing between induction of transcription and the appearance of a specific hybridization signal with the probes complementary to the 3' ends of the transcripts (Fig.1) was determined by dot blot hybridization, as described in Materials and Methods." (BNID 104900 measurement section:) "The mRNA chain elongation rate was determined by the time lag between induction of transcription of specific mRNAs (lacZ, infB) and the appearance of specific hybridization to DNA probes from the 3' ends of the respective genes." |
Comments |
Slowest rate 28 nts/sec on glucose-15 alpha-methyglucoside medium (doubling time 200 min) fastest rate 89 nts/sec on glucose casamino acid medium (doubling time 35 min) |
