Method Chloroplasts from spinach leaves that were homogenized in an isolation medium were precipitated precipitated by centrifugation and suspended in the medium. Measurement of Cytochrome c reduction induced by a single turnover flash.
Comments The following assumptions were made: (1) All O2- produced was trapped by Cyt c. (2) The reaction: [O2-(out)+oxidized Cyt c>>O2+reduced Cyt c] was a unidirectional flux because the O2- concentration was diluted by thousands of times in the bulk suspension medium. (3) The rate constant of above reaction was 5E5/(M *SEC) AT pH 7.5
