Method Abstract: "Here [investigators] report the crystal structure of the gap junction channel formed by human connexin 26 (Cx26, also known as GJB2) at 3.5 A resolution, and discuss structural determinants of solute transport through the channel." P.601 right column bottom paragraph: "Methods summary: Human Cx26 was expressed in Sf9 insect cells using recombinant baculovirus. The gap junction channel was solubilized in dodecylmaltoside and purified sequentially by cation exchange and size-exclusion chromatography. Crystals were grown by the hanging-drop vapour diffusion method with PEG200 as a precipitant. The structure was determined by the single-isomorphous replacement combined with anomalous scattering (SIRAS) method, with phase extension by six-fold non-crystallographic (NCS) averaging."
