P.885 right column bottom paragraph: "To complement [investigators'] flow cytometric and microscopic analyses that defined the quantitative distribution of human ES cells in different cell cycle phases, [they] directly measured the duration of the human ES cell cycle. [They] determined the rate of human ES cell proliferation by pulse labeling with CFSE [Carboxy-fluorescein diacetate succinimidyl ester]. CFSE attaches non-specifically to cellular proteins and its stochastic distribution among progeny cells provides a measure of cell division. [They] followed the amount of CFSE autofluorescence per cell by FACS [Fluorescence-Activated Cell Sorting] as a function of time over the course of 3 days. This approach defines the distribution of cells labeled with different fluorescence intensities relative to the total number of cells at the time of labeling (Fig. 3)." |

Because most actively proliferating somatic cells require ∼24–32 h to complete cell division, the CFSE incorporation data clearly demonstrate that human ES cells have an abbreviated cell cycle." |
