Method "O2 evolution measurements were performed with a Clark-type 02 electrode (Rank Brothers, Cambridge UK). The samples were maintained at 30°C in a water jacketed cuvette. A 500 W light source was used to illuminate the sample at a light intensity (1200µmoles/m^2/sec) that saturated photosynthesis."
Comments "The light-saturated rate of O2 evolution declined dramatically in cells deprived of S for 48 h and was reduced by about 40% in cells deprived of P for the same period (Table 2)...The ES [energy storage] capacities presented in Table 2 represent energy storage per unit of sample volume. The volume of the sample that is measured in the photoacoustic technique, as applied here, can be affected by three parameters of the sample: its thickness, its optical density at the wavelength of the modulated light and its thermal diffusion properties (Carpentier et al. 1983 Tam 1986 Buschmann and Prehn 1990)." Was unable to determine how cells were counted in this study.
