Method "Characterization of the cell volume following fusion. Cells were fused with DOPC GUVs as in Fig. 2, followed by fixation and phalloidin staining. For each cell, spinning-disk confocal stacks were taken and the 3D cell outline was determined by segmenting the cell based on the fluorescence images. The cell volume was taken as a sum of the cell body volume and the lamellipodial volume which was approximated as the product of the lamellipodial area and its thickness (h~200 nm). Histograms of the cell volume distributions in a population of fixed cells after fusion and a control population of cells from the same samples which had not undergone fusion are shown. The population mean and standard deviation are illustrated above the histograms. Fused cells have similar volume to control cells (p>0.1)."
