Method "Using transgenic mice harboring a chromosomally integrated lacZ reporter gene...To capture most of the epithelial cells, [researchers] scraped off the innermost layer of the intestine (mucosa) with the surface villi. Figure 5A shows the intact mucosa. [They] then determined the LacZ mutant frequencies in the mucosal prep (Figure 5B) and in the remaining fraction, which consists of the serosa, muscularis and part of the lamina propia with some crypts still visible (Figure 5C). For brevity [they] call this the serosa throughout the text. These two fractions were analyzed for the duodenum, jejunum and ileum from 3 to 6 mice per determination point at 19 months (Figure 6A, Table S2)."
