"The growth medium was either Medium C
(Helmstetter, 1967), supplemented with 0.2% (v/v) of
glycerol and the required amino acids at 50 mg/ml, or LB
medium (Miller, 1972) supplemented with 0.2%
(w/v) of glucose. Experimental cultures were grown in a
shaker waterbath at 37°C after inoculation from overnight
cultures in glycerol minimal medium, either 1:250 into the
same medium or 1:2000 into LB medium. Growth was followed
by measuring the optical density at 600 nm wavelength
and 1 cm light path (A600)." |
