Method "Oxygen equilibrium curves were measured as described by Imai et al. (1970). Measurements were carried out on purified D. akajei Hb at 298 K in 50 mM 2-[bis(2-hydroxymethyl) amino]-2-hydroxymethyl-1,3-propanediol (bis-Tris) or Tris buffer with 100 mM chloride.In order to reduce methaemoglobin levels, catalase and superoxide dismutase were added to Hb samples (Lynch et al., 1976 Winterbourn et al., 1976). Deoxygenation curves were used to determine p50 (the oxygen partial pressure necessary to attain 50% saturation of Hb with oxygen) and the Hill coefficient n (the maximum slope of Hill plots of oxygen equilibrium curves). 2 mM ATP was used as an organic phosphate effector."
Comments "The oxygen equilibrium characteristics are presented in Table 2. D. akajei Hb shows a slightly lower oxygen affinity than that of human HbA, with moderate haem-haem interaction, Bohr effect and ATP effect (Imai, 1982)." See note beneath table
