Method "According to classic reports from the Krebs' laboratory [refs 35,36] and other studies, [ref 37] direct measurement of NAD, NADH, NADP, and NADPH is not informative in terms of control of metabolism because they do not provide information on compartmentalization of nicotinamide nucleotides between cytosol and mitochondria and because they do not separate free from protein-bound forms (only free fractions determine direction and free-energy changes of dehydrogenase reactions). The same studies proposed an alternative approach, implying assessment of free NAD(P)+/NAD(P)H ratios in the cytoplasm and mitochondria by measuring the ratios of the concentrations of the oxidized and reduced metabolites of suitable NAD(P)-linked dehydrogenase systems."
